In Australia, in the past, pasture legumes were rotated mainly with cereals, but increasingly these rotations now involve pasture legumes with a wider range of crops, including legumes. This increasing frequency of the leguminous host in the rotation system may be associated with increased root rots in legumes in the current pasture-crop rotations. The primary aim of this study was to see whether the pathogenicity on pasture legumes of strains of Rhizoctonia solani sourced from lupins and cereals (common crops in rotation with pastures) is associated with increased incidence of root rots in pasture legumes in the disease conducive sandy soils of the Mediterranean regions of southern Australia. The second aim was to determine sources of resistance among newly introduced pasture legumes to R. solani strains originating from rotational crops as this would reduce the impact of disease in the pasture phase. Fifteen pasture legume genotypes were assessed for their resistance/susceptibility to five different zymogram groups (ZG) of the root rot pathogen R. solani under glasshouse conditions. Of the R. solani groups tested, ZG1–5 and ZG1–4 (both known to be pathogenic on cereals and legumes) overall, caused the most severe root disease across the genotypes tested, significantly more than ZG6 (known to be pathogenic on legumes), in turn significantly >ZG4 (known to be pathogenic on legumes) which in turn was >ZG11 (known to be pathogenic on legumes including tropical species). Overall, Ornithopus sativus Brot. cvs Cadiz and Margurita, Trifolium michelianum Savi. cvs Paradana and Frontier and T. purpureum Loisel. cv. Electro showed a significant level of resistance to root rot caused by R. solani ZG11 (root disease scores ≤1.2 on a 1–3 scale where 3 = maximum disease severity) while O. sativus cvs Cadiz and Erica showed a significant level of resistance to root rot caused by R. solani ZG4 (scores ≤1.2). O. compressus L. cvs Charano and Frontier, O. sativus cv. Erica, and T. purpureum cv. Electro showed some useful resistance to root rot caused by R. solani ZG6 (scores ≤1.8). This is the first time that cvs Cadiz, Electro, Frontier, Margurita and Paradana have been recognised for their levels of resistance to root rot caused by R. solani ZG11; and similarly for cvs Cadiz and Erica against ZG4; and for cvs Charano, Erica, and Electro against ZG6. These genotypes with resistance may also serve as useful sources of resistance in pasture legume breeding programs and also could potentially be exploited directly into areas where other rotation crops are affected by these R. solani strains. None of the tested genotypes showed useful resistance to R. solani ZG1–4 (scores ≥2.0) or ZG1–5 (scores ≥2.5). This study demonstrates the relative potential of the various R. solani ZG strains, and particularly ZG1–4, ZG1–5, ZG4 and ZG6 to attack legume pastures and pose a significant threat to non-pasture crop species susceptible to these strains grown in rotation with these pasture legumes. Significantly, the cross-pathogenicity of these strains could result in the continuous build-up of inoculum of these strains that may seriously affect the productivity eventually of legumes in all rotations. In particular, when choosing pasture legumes as rotation crops, caution needs to be exercised so that the cultivars deployed are those with the best resistance to the R. solani ZGs most likely to be prevalent at the location.  相似文献

Populations of viable sclerotia ofSclerotium rolfsii were highest in soil in a field in which tomato was planted for three successive years before sampling and in one in which tomato followed groundnut in the 2 years prior to this study. Summary We report the complementation of a genetic lesion in the genome of Erwinia carotovora subsp. carotovora (Ecc), a pathogenic bacterium that incites soft rot of plants. A Sau3AI genomic library of Ecc was constructed using the conjugal cosmid pLAFR-3 as a vector. Sixteen cosmid clones encoding various plant tissue-degrading enzymes were identified, including a proteolytic clone, five cellulolytic clones, and ten pectolytic clones. We detected a mutant of Ecc with no proteolytic activity following transposon mutagenesis with an unstable Tn5-carrying plasmid. Conjugal transfer of the protease-encoding cosmid to this mutant restored near-wildtype extracellular protease production. Further manipulation and study of genes encoding pathogenic determinants in Ecc will be possible using this system.  相似文献

The interactions of Heterodera glycines at four egg inoculum levels (0, 100, 1,000, and 10,000 per pot) and three cyst levels (0, 100, and 200 per pot) and Calonectria crotalariae at 500, 5,000, and 50,000 microsclerotia per pot were evaluated on soybean. At the two lowest nematode egg levels, the presence of C. crotalariae did not affect nematode reproduction. At 10,000 eggs per pot, however, nematode reproduction was increased significantly at each microsclerotial level. The increase in nematode reproduction was stepwise at 500 and 5,000 microsclerotia per pot but declined at 50,000 microsclerotia per pot. Similar results were obtained when cysts rather than eggs were used as nematode inoculum. The nematode x fungus interaction significantly affected 60-day plant growth parameters of both Lee 74 and Centennial soybean. The nematode x fungus interaction was antagonistic to plant roots and significantly influenced root injury ratings. The presence of C. crotalariae in tissues of stock plants or plants used as race differentials did not alter the analysis of this population as race 3.  相似文献

Summary Quantitative inheritance of resistance to Phytophthora pod rot (Ppr) was studied in cocoa hybrid progeny from 12 Trinitario x Amazonian crosses and their reciprocal crosses. The crossing scheme was similar to a factorial design. Disease was assessed by the number and percentage of infected pods on each tree. Highly significant differences due to general combining abilities (GCA) were obtained for all characters, except for the GCA of Trinitario on total pod production. Differences for specific combining ability (SCA) were not significant for all characters. There were no significant differences between reciprocal crosses. The Trinitario clone K82 provided the only source for the hybrid progenies of strong Ppr resistance to the hybrid progenies, while K20 provided moderate resistance. Other parental clones — KA2-101, KA5-201, KEE 2, KEE 5, and KEE 52 — produced progenies which were susceptible to Ppr. It is evident that resistance to Ppr in cocoa is inherited additively. Maternal and cytoplasmic effects were assumed to have no influence on inheritance of resistance. It is also concluded that resistance to Ppr of the kind shown by K82 is likely to be horizontal resistance. Breeding for high-yielding cultivars combined with Ppr resistance is the most effective way of controlling Ppr of cocoa on the crops of growers with small holdings in Papua New Guinea.  相似文献

Abstract Using fungi grown on synthetic agar medium, we evaluated and compared the concentration of various H2O2-producing enzymes. Our results showed that oxidase production in solid medium was better than that found in liquid medium and as high as that detected in wood samples. High yields of oxidases made it possible to compare different oxidases in the same culture extracts and under different conditions. Our results also indicated that H2O2 production is ubiquitous in the white rot fungi tested and that enzyme levels are influenced by the substrate composition.  相似文献

A crown rot disease in wheat caused by the fungusFusarium graminearum Schw. Group 1 is a widespread problem in chronically Zn-deficient Australian soils. A link between crown rot and Zn deficiency was established by Sparrow and Graham (1988). This paper reports a test of a further hypothesis, that wheat genotypes more efficient at extracting zinc from low-zinc soils are more resistant to infection by this pathogen. Three wheat cultivars (Excalibur, Songlen and Durati) of differential Zn efficiency were tested at three zinc levels (0.05, 0.5 and 2.0 mg Zn kg−1 of soil) and three levels ofF. graminearum S. Group 1 inoculum (0.1 g and 0.3 g kg−1 live chaff-inoculum and control having 0.1 g kg−1 dead chaff inoculum). Six weeks after sowing dry matter production of shoots and roots was decreased byFusarium inoculation at 0.05 mg and 0.5 mg kg−1 applied Zn.Fusarium inoculum at 0.1 g was as effective as 0.3 g kg−1 for infection and decreasing dry matter. The infection at the basal part of culm decreased significantly by increasing the rate of Zn application. Excalibur, a Zn-efficient cultivar (tolerant to Zn deficiency) produced significantly more shoot and root dry matter, and showed less disease infection compared with Zn-inefficient cultivars (Durati and Songlen) at low (0.05 mg Zn kg−1 soil) and medium (0.5 mg Zn kg−1 soil) Zn fertilization rates. Higher rate of Zn fertilization (2.0 mg Zn kg−1 soil) reduced the disease level in Durati to the level of Excalibur but the disease level of Songlen was still high, indicating its high Zn requirement and or sensitivity to crown rot. The data on Zn uptake show that Excalibur, being Zn-efficient, was able to scavenge enough Zn from Zn-deficient soil, we suggest that besides sustaining growth Excalibur was able to build and maintain resistance to the pathogen; inefficient cultivars needed extra Zn fertilization to achieve performance comparable to that of Excalibur. The present study indicates that growing Zn-efficient cultivars of wheat along with judicious use of Zn fertilizer in Zn-deficient areas where crown rot is a problem may sustain wheat production by reducing the severity of the disease as well as by increasing the plant vigour through improved Zn nutrition. ei]Section editor: R Rodriques-Kalana  相似文献

Strawberry tissues infected with Phytophthora cactorum were comminuted and plated in a selective antibiotic agar medium to determine levels of tissue colonisation as indicated by the number of colony forming units (CFU) recovered per gramme of infected tissue. The number of CFU recovered per gramme of tissue increased logarithmically with the amount of necrosis in infected crown, leaf and petiole tissues. Under the conditions of enhanced susceptibility to infection and colonisation caused by cold storage treatments, this relationship between colonisation and necrosis was not significantly altered in the susceptible cv. Tamella. A recovery index was used to determine the effect of infected tissues on the recovery of CFU. This indicated that increasing levels of host colonisation stimulated CFU recovery and may partly explain the large increase in CFU g-1 with larger amounts of necrosis. The amount of tissue colonisation was greater in inoculated plants of the susceptible cv. Tamella than in less susceptible cv. Cambridge Favourite, although the necrotic tissues of the latter contained more CFU g-1, indicating a greater level of tolerance to colonisation. In cv. Tamella small amounts of colonisation were capable of causing wilt symptoms, although no wilted plants contained less than 200 CFU g-1. Conversely, plants containing more than 1000 CFU g-1 always wilted. In the early stages of infection, low levels of colonisation could be detected in strawberry crowns in the absence of symptoms. Dormant strawberry plants of cv. Tamella were readily infected by P. cactorum zoospore inoculations but, unlike actively growing plants, the majority of infections remained latent. These latent infections exhibited little or no symptoms and CFU recoveries from infected tissues were always below 100 CFU g-1.  相似文献

The severity of a root rot disease of cereals, caused by Rhizoctonia solani Kühn AG8, was inversely correlated to the Zn status of plants in field studies in 1989 and 1990. In 1989, a preliminary survey was conducted in a farmer's field in South Australia where Zn deficiency and disease were both widespread. Zn concentration in Spear wheat plants at the 3-leaf to early tillering stage was negatively correlated with severity of the disease. For the elevent elements analysed, a correlation matrix showed that Zn had the highest, and only significant (R2=0.52**) association with disease. The effect of Zn applications and their residual value on disease severity was further studied in a long-term field experiment in 1989 and 1990 to which Zn had been applied in 1986. There was a decrease in the area of Rhizoctonia bare patch as Zn rate was increased, a result consistent with the field survey results; the recommended rate of 2.5 kg Zn ha–1 reduced the area affected by bare patch from 42% to 21% of the total crop area compared with no Zn application, overcame Zn deficiency and increased grain yield from 1.1 to 2.8 t ha–1. In 1990, fresh Zn application treatments were applied to trial plots designed for this purpose, in order to compare the response with the older Zn treatments applied in 1986. The areas of bare patch in the older Zn treatments were approximately 5% greater than those in the fresh Zn treatments. The results are consistent with the hypothesis that Zn deficient plants are more susceptible to root rot caused by R. solani. Testing this hypothesis is the subject of a companion paper.  相似文献

Root rot of pawpaw (Carica papaya L.) reported in Nigeria is caused byPythium aphanidermatum which was consistently isolated from diseased plant parts and highly pothogenic. Out of 16 different media tested, it grew best on corn-meal-agar (CMA) and CMA supplemented with cellulose and sucrose. The highest number of oospores/ml was on CMA with average diameter of 19.9±0.1 µm. The symptom is characterized by dark brown rot of roots, absence of secondary roots and disintegration of internal tissue of the main root. These cause the progressive decline of the aerial parts of the tree untill it dies.  相似文献
